TREATMENT WITH HYPERTHERMIA IN MURINE BREAST CANCER CELLS

UBreast cancer is one of the most common and serious diseases, especially among the female population, and is currently considered a worldwide public health problem. Although conventional treatments such as chemotherapy and radiotherapy have good results, it remains a challenge to minimize their side effects. An alternative cancer treatment is hyperthermia. This technique aims to eliminate cancerous cells, which undergo heat for more than 30 minutes coagulate the nucleus. This work aimed to evaluate the cell death of murine breast cancer cells (Erlich tumor) after being submitted to hyperthermia. For this study we used 1×105 murine breast carcinoma cells (ATCC® CCL-77TM). Cells were subjected to high temperatures (40C°, 42C°, 44C°, 46C°, 48C°, 50C° and 60C°) by means of a thermostatic bath for 30 minutes. The treatments were done in triplicates and evaluated by Trypan Blue Method and Flow Cytometry (Annexin V-FITC / Propidium Iodide Assay). The temperature that inhibited 50% of the cell viability determined by the curve was analyzed using the statistical program GraphPad Prism 4. The temperature which inhibited 50% cell viability was 48 ° C, which had a statistically significant difference with the 50 ° C temperature which inhibited more than 70% viability. Through flow cytometry it was also possible to identify that the most predominant cell death was necrosis. Based on the results obtained it can be inferred that hyperthermia causes a considerable drop in the cell viability of murine breast cancer cells. So it is a very promising study in the fight against breast cancer. Because, besides contributing as a new efficient and safe antitumor treatment, it can be associated with chemotherapy and potentiate its efficiency.Ehrlich ascites carcinoma
