EFFECTS OF MILK COLLECTION TIME ON THE FUNCTIONAL ACTIVITY OF COLOSTRUM PHAGOCYTES IN THE PRESENCE OF NANOFRACTIONS OF MELATONIN ADSORBED TO THE PEG MICROSPHERE.

Federal University of Mato Grosso (UFMT); Federal University of Mato Grosso do Sul (UFMS).The aim of this study was verified the effects of milk collection time (diurnal/nocturnal) on the functional activity of colostrum phagocytes in the presence of nanofractions of melatonin adsorbed to the PEG (polyethylene glycol) microsphere. Colostrum samples were collected from 30 mothers aged between 18 and 40 years during the day and night, for a total of 60 samples. The cells were incubated at 37°C for 24 h in a humid atmosphere containing 5% CO2 (carbon dioxide) in the presence or absence of melatonin adsorbed onto the PEG microsphere. After this period the culture supernatant was removed and reserved for CuZn-SOD (enzymatics of dismutase superoxide) quantification by the NBT (nitrotetrazolium blue) reduction method. Cells were resuspended in cytochrome C to verify superoxide anion. It was observed, regardless of the treatment and phase of the day, there was no difference in CuZn-SOD concentration in the culture supernatant. The superoxide release was higher in colostrum samples collected at night. Melatonin, adsorbed or not in PEG microsphere, reduced the superoxide release by MN phagocytes from diurnal samples. These data suggest that melatonin may act as an antioxidant in the daytime phase, whereas in the night phase it may exert prooxidative effects on colostrum cells.breast cancer
