EVALUATION OF ANTICANCER ACTIVITY OF LYCOPENE RICH EXTRACT FROM RED GUAVA (PSIDIUM GUAJAVA L.) IN HUMAN BREAST ADENOCARCINOMA CELL LINE MCF-7.

Universidade de BrasíliaThe aim of the study was to investigate the potential activity of lycopene extract from guava (LEG) in human breast adenocarcinoma (HBA) cell line MCF-7, in vitro. Firstly, LEG rate of 20% was obtained from 500 g of fresh guava using organics solvents. The characterization and quantification of lycopene in the sample were performed by UV-Vis and ATR-FTIR. The viability of cells was analyzed by colorimetric assay MTT after 24 and 72 hours of exposure to several concentrations (6.25 to 1.600 µg/mL). The flow cytometry was performed to investigate the which possible mechanism involved in the process of cells death. Therefore, DNA fragmentation and cell cycle assay were realized using propidium iodide as marker of nucleic acid. After times exposure, it was possible to observe a dose and time-dependent activity of LEG in MCF-7 viability. The IC50 was approximately of 50 and 6.25 µg/mL after 24 and 72 hours, respectively. The proliferation of cells was significantly reduced by 80% after 72 hours of exposure and no membrane injury was noted. Moreover, the cells exposed to LEG presented an increase of DNA fragmented of 50 and 60% after 24 and 72 hours of exposure, respectively. In parallel, the cell cycle was disturbed by significant increase (20%) of cells in phase G1 and reduction (10%) of cells in phase S after 72 hours of exposure. These results showed that LEG has potential to reduce the viability and proliferation of MCF-7 cells line and generates DNA fragmentation and cell cycle stop in vitro. Therefore, LEG demonstrated potential anticancer activity against HBA, in vitro. However, more experiment will be performed to understand better the mechanism of cells death.flow cytometry, Lycopene, MCF-7, Psidium guajava
